目的 研究实验性肝纤维化过程中明胶酶A(mmp-2)酶原蛋白表达的动态变化。 方法 建立四氯化碳大鼠肝纤维化模型,采用Western印迹法和免疫组化方法,对肝纤维化不同阶段肝内mmp-2的酶原蛋白的表达水平与分布进行了检测。 结果 在正常大鼠肝组织中有mmp-2的酶原表达。在肝纤维化发生发展过程中mmp-2酶原蛋白表达水平逐渐增强,至肝纤维化中期到达最高峰,到肝纤维化晚期及肝硬变阶段其表达较中期下降,但仍高于正常对照水平。正常肝组织内,mmp-2的表达主要位于汇管区的胆管壁和毛细血管壁及肝窦壁的间质细胞;纤维化时,增生的纤维间隔附近亦有较高表达。 结论 mmp-2在肝纤维化的发生、发展直至肝硬变形成过程中发挥着重要作用。
Objective To study the dynamic changes of zymogram protein (MMP-2) in experimental hepatic fibrosis. Methods The model of liver fibrosis induced by carbon tetrachloride was established in rats. Western blotting and immunohistochemistry were used to detect the expression of mmp-2 mRNA and protein in different stages of hepatic fibrosis. Results The expression of mmp-2 was found in normal rat liver tissue. During the development of hepatic fibrosis, the expression of mmp-2 proenzyme increased gradually, reaching its peak in the middle stage of liver fibrosis, and decreased to the stage of advanced stage of liver fibrosis and cirrhosis, but still higher than that of the normal control Level. In normal liver tissue, the expression of mmp-2 mainly located in the bile duct wall, the capillary wall and the hepatic sinusoidal wall of interstitial cells in the portal area. When fibrosis, the hyperplastic fibroids also had a high expression in the vicinity of the fiber space. Conclusion mmp-2 plays an important role in the development of hepatic fibrosis and cirrhosis.
