为了评价低温条件下东北林蛙的代谢规律和乳酸脱氢酶(LDH)酶谱特点,试验采用垂直板聚丙烯酰胺凝胶电泳法和紫外吸收法,分别对东北林蛙不同组织的同工酶酶谱特点和酶活力进行分析。结果表明:LDH同工酶的分布和含量有明显的组织特异性,肝脏、肾脏、心脏和肌肉均缺失LDH2;肝脏和肌肉中LDH5同工酶相对含量高于LDH4,LDH1和LDH3缺失;肾脏中LDH5>LDH1>LDH4,LDH3缺失;心脏中LDH1>LDH3≈LDH4>LDH5,4℃组和18℃组特点相同。4℃组与18℃组相比,低温状态下东北林蛙处于较低的糖代谢水平,肝脏、肾脏、心脏和肌肉中LDH总酶活力分别降低了12.2%、36.5%、32.5%、15.0%,各同工酶相对含量均有不同程度降低。
In order to evaluate the metabolic regularity and lactate dehydrogenase (LDH) zymogram of Rana chensinensis under low temperature conditions, the vertical plate polyacrylamide gel electrophoresis and UV absorption were used to analyze the isozymes Zymogram characteristics and enzyme activity were analyzed. The results showed that the distribution and content of LDH isoenzyme had obvious tissue specificity, LDH2 was lost in liver, kidney, heart and muscle; the relative content of LDH5 isoenzyme in liver and muscle was higher than that of LDH4 and LDH1 and LDH3; LDH5> LDH1> LDH4, LDH3 deletion; heart LDH1> LDH3 ≈ LDH4> LDH5, 4 ℃ group and 18 ℃ group the same characteristics. Compared with the 18 ℃ group, 4 ℃ group had lower glucose metabolism level at low temperature, and the total LDH activity of liver, kidney, heart and muscle decreased by 12.2%, 36.5%, 32.5% and 15.0% , The relative content of isozymes were reduced to varying degrees.
