目的探讨垂体腺瘤中半乳糖凝集素-3(Galectin-3,Gal-3)和凋亡相关基因Bcl-2、Bax的内在联系。方法RT-PCR检测20例垂体腺瘤Gal-3、Bcl-2和BaxmRNA的表达;免疫组化染色检测78例垂体腺瘤中Gal-3、Bcl-2和Bax蛋白表达,分析侵袭性和非侵袭性垂体腺瘤中Gal-3、Bcl-2和Bax表达的差异及其相关关系。结果侵袭组垂体腺瘤Gal-3和Bcl-2mRNA和蛋白的表达水平较非侵袭组显著增高,而BaxmRNA和蛋白在两组中的表达水平无显著差异。等级相关分析显示Gal-3蛋白与Bcl-2蛋白之间有正直线相关关系(r=0.291,P=0.01);Gal-3蛋白与Bax蛋白之间无直线相关关系(r=-0.023,P>0.05);Bcl-2蛋白与Bax蛋白表达之间的Pearson相关系数为r=-0.267,P<0.05(双侧),呈负相关关系。结论Gal-3可能主要是通过与Bcl-2协同作用,发挥其抗凋亡效应,相对促进垂体腺瘤细胞增殖,导致垂体腺瘤的侵袭性生长。这些结果提示Gal-3在垂体肿瘤细胞增殖过程中发挥重要作用,有望作为垂体腺瘤的一个治疗靶点。
Objective To investigate the intrinsic relationship between Galectin-3 (Gal-3) and Bcl-2 and Bax in pituitary adenomas. Methods The expressions of Gal-3, Bcl-2 and Bax mRNA in 20 pituitary adenomas were detected by RT-PCR. The protein expressions of Gal-3, Bcl-2 and Bax in 78 pituitary adenomas were detected by immunohistochemical staining. The difference of the expression of Gal-3, Bcl-2 and Bax in invasive pituitary adenomas and its correlation. Results The expression of Gal-3 and Bcl-2 mRNA and protein in pituitary adenomas of invasive group was significantly higher than that of non-invasive group, but the expression of Bax mRNA and protein in the two groups had no significant difference. There was a positive correlation between Gal-3 protein and Bcl-2 protein (r = 0.291, P = 0.01). There was no linear correlation between Gal-3 protein and Bax protein (r = -0.023, P > 0.05). The Pearson correlation coefficient between Bcl-2 protein and Bax protein was negative (r = -0.267, P <0.05). Conclusions Gal-3 may exert its anti-apoptotic effect mainly through the synergistic effect with Bcl-2, which may promote the proliferation of pituitary adenoma cells and lead to the invasive growth of pituitary adenomas. These results suggest that Gal-3 plays an important role in the proliferation of pituitary tumor cells and is expected to serve as a therapeutic target for pituitary adenomas.
