目的:观察美丽鳞毛蕨粗多糖对α-葡萄糖苷酶抑制活性的影响及对蔗糖、葡萄糖、麦芽糖、淀粉负荷剂量后的正常小鼠血糖影响。方法:采用α-葡萄糖苷酶活性测定方法,对美丽鳞毛蕨植物粗多糖进行α-葡萄糖苷酶抑制活性的测定;以美丽鳞毛蕨粗多糖为实验药物,取血糖值4.0~9.0 mmol·L-1的小鼠,按体重随机分成16小组,每组10只,适应性饲养3 d后,分别用生理盐水、阿卡波糖(0.625 g·kg-1)、高剂量粗多糖(0.48 g·kg-1)、低剂量粗多糖(0.24 g·kg-1)ig小鼠,每天1次,连续ig 4 d造模;第5 d分别用葡萄糖(2.0 g·kg-1)、蔗糖(4.0 g·kg-1)、麦芽糖(4.0 g·kg-1)、淀粉(6.0 g·kg-1)给小鼠负荷剂量,在15,30,60,120 min时间点测量小鼠血糖值并记录。观察美丽鳞毛蕨粗多糖对蔗糖、葡萄糖、麦芽糖、淀粉负荷剂量后的正常小鼠血糖影响。结果:美丽鳞毛蕨粗多糖在样品浓度10 g·L-1反应体系下,对α-葡萄糖苷酶抑制活性达到90.8%以上,属于竞争性抑制类型(Ki=2.53 g·L-1);以美丽鳞毛蕨粗多糖为实验药物,淀粉蔗糖负荷剂量后小鼠耐糖量与对照组比较,淀粉组和麦芽糖组血糖值降低较为明显(P<0.01),蔗糖组次之(P<0.05)。结论:美丽鳞毛蕨粗多糖对α-葡萄糖苷酶抑制活性高,属于竞争性抑制,并能提高正常小鼠的耐糖量。
OBJECTIVE: To observe the effect of the polysaccharides from the genus Pholiota on the inhibitory activity of α-glucosidase and the effects on the blood glucose of normal mice after sucrose, glucose, maltose and starch loading. Methods: The α-glucosidase inhibitory activity of crude polysaccharides from Pleurotus ostreatus was determined by measuring the activity of α-glucosidase. The polysaccharides of Pleurotus ferulae were used as experimental drugs, and the blood glucose level was 4.0-9.0 mmol · L-1 mice were randomly divided into 16 groups according to body weight, with 10 mice in each group. After three days of adaptive feeding, the mice were treated with physiological saline, acarbose (0.625 g · kg-1), high dose of crude polysaccharide (ig) and low dose of crude polysaccharides (0.24 g · kg-1) once a day for 4 days. The rats were sacrificed on the 5th day with glucose (2.0 g · kg -1), sucrose (4.0 g · kg-1), maltose (4.0 g · kg-1) and starch (6.0 g · kg-1) were given to mice. The blood glucose of mice was measured at 15, 30, 60, . To observe the effect of the polysaccharides of Pleurotus ferulae on the blood glucose of normal mice after sucrose, glucose, maltose and starch loading. Results: The inhibitory activity of α-glucosidase was more than 90.8% under the concentration of 10 g · L-1, which was competitive inhibition type (Ki = 2.53 g · L-1). Compared with the control group, the blood sugar level of starch group and maltose group was significantly decreased (P <0.01), sucrose group was the second (P <0.05) . CONCLUSION: The polysaccharides of Pseudostellariae spectabilis have high inhibitory activity on α-glucosidase and are competitive inhibition, and can increase the sugar tolerance of normal mice.
