目的:探讨扶正固本中药对内毒素所致大鼠急性肺损伤(Acute Lung Injury)的免疫调节作用。方法:30只SD雄性大鼠随机分成5组:正常对照(NS)组、急性肺损伤(ALI)组、黄芪(HQ)组、参附(SF)组、黄芪+参附(HQ+SF)组。NS组、ALI组予以生理盐水(NS)腹腔注射,HQ组予以黄芪注射液腹腔注射,SF组予以参附注射液腹腔注射,HQ+SF组予以黄芪注射液+参附注射液腹腔注射。给药7天后除正常对照组外均尾静脉注射内毒素。分别测定各组大鼠支气管肺泡灌洗液(BALF)及血液中干扰素-γ(IFN-γ)、白介素-4(IL-4)浓度。同时观察各组大鼠肺组织的病理改变。结果:(1)与正常对照组比较,ALI组大鼠BALF及血液中IFN-γ含量明显减少(P<0.01),IL-4含量明显增加(P<0.01)。与ALI伤组比较,HQ组IFN-γ含量明显增加(P<0.01),SF组IFN-γ含量无明显变化(P>0.05),HQ+SF组IFN-γ含量较ALI组、HQ组、SF组明显增加(P<0.01)。而各组IL-4含量较ALI组无明显变化(P>0.05)。(2)与ALI组相比,HQ组、SF组、HQ+SF组的肺系数、BALF中的蛋白含量均低于ALI组,其中HQ+SF组下降更为明显。结论:黄芪、参附、黄芪加参附中药液对ALI均有防治作用。其中黄芪、黄芪加参附中药液通过调节炎症反应而发挥对ALI的防治作用,对ALI以促炎症反应为主时可能更为有益;参附对抗炎症介质和促炎症介质均无明显影响,但肺病理结果显示肺损伤程度明显减轻,其对ALI的防治作用可能通过其他途径实现。
Objective: To investigate the immunomodulatory effects of Fuzheng Guben Chinese Herbs on endotoxin-induced acute lung injury (Acute Lung Injury) in rats. Methods: Thirty SD male rats were randomly divided into 5 groups: normal control (NS), acute lung injury (ALI), Astragalus membranaceus (HQ), Shenfu (SF) group. The rats in NS group and ALI group were intraperitoneally injected with NS. The rats in HQ group were given intraperitoneal injection of Astragalus injection, the rats in SF group were injected intraperitoneally with Shenfu injection, the mice in HQ + SF group were injected intraperitoneally with Astragalus injection and Shenfu injection. After 7 days of administration, endotoxin was injected into the tail vein except the normal control group. The concentrations of interferon-γ (IFN-γ) and interleukin-4 (IL-4) in bronchoalveolar lavage fluid (BALF) and blood were measured in each group. At the same time observe the pathological changes of lung tissue in each group. Results: (1) Compared with normal control group, the content of IFN-γ in BALF and blood in ALI group was significantly decreased (P <0.01) and IL-4 content was significantly increased (P <0.01). Compared with ALI group, the content of IFN-γ in HQ group was significantly increased (P <0.01), the level of IFN-γ in SF group was not significantly changed (P> 0.05) SF group increased significantly (P <0.01). The content of IL-4 in each group had no significant change compared with ALI group (P> 0.05). (2) Compared with ALI group, the lung coefficient and the protein content of BALF in HQ group, SF group and HQ + SF group were lower than those in ALI group, especially in HQ + SF group. Conclusion: Astragalus, ginseng, Astragalus and ginseng with Chinese medicine liquid have preventive and therapeutic effects on ALI. Among them, astragalus, astragalus and ginseng with Chinese medicine liquid can play a role in prevention and treatment of ALI by regulating inflammatory reaction, and may be more beneficial to ALI with predominantly proinflammatory reaction. There is no obvious effect of ginseng on anti-inflammatory mediators and proinflammatory mediators Lung pathology results showed that the degree of lung injury was significantly reduced, its prevention and treatment of ALI may be achieved by other means.
