为准确地在体内评价Wistar大鼠的胰岛素敏感性,我们建立了大鼠的正常血糖-高血浆胰岛素钳夹技术(简称钳夹技术).并运用该技术初步探讨了年龄,体重、血浆胰岛素及血脂水平对胰岛素敏感性的影响。结果显示,我们建立的钳夹技术所测定的葡萄糖输注率(GIR)、细胞外液变化量及其它指标均接近或优于文献报道。同时发现,年龄、体重大于Ⅰ组大鼠的Ⅱ组大鼠(P<0.01).其GIR、血浆高密度脂蛋白组分2水平显著低于Ⅰ组大鼠(P<0.01).而血浆胰岛素及血浆甘油三酯水平显著高于Ⅰ组大鼠(P<0.01和0.05)。本研究的结果提示:(1)我们建立的钳夹技术较准确、可靠(2)随着年龄、体重的增大,大鼠的胰岛素敏感性逐渐下降。
To accurately assess insulin sensitivity in Wistar rats in vivo, we established a normal glucose-high plasma insulin clamp technique (abbreviated as clamp technique) in rats. The effects of age, body weight, plasma insulin and serum lipids on insulin sensitivity were also discussed. The results showed that the glucose infusion rate (GIR), extracellular fluid changes and other parameters measured by the clamp technology we established were close to or better than those reported in the literature. At the same time, it was found that the age and weight of rats in group Ⅱ were significantly higher than those in group Ⅰ (P <0.01). The levels of GIR and plasma high density lipoprotein fraction 2 were significantly lower than those in group I (P <0.01). Plasma insulin and plasma triglyceride levels were significantly higher than those in group I (P <0.01 and 0.05). The results of this study suggest that: (1) The clamp technique we established is more accurate and reliable. (2) As the age and body weight increase, the insulin sensitivity of rats gradually decreases.
