目的 :观察早期糖尿病大鼠血清对培养视网膜M櫣ller细胞 (retinalM櫣llercells,RMCs)增殖的影响 .方法 :将链脲霉素 (streptozotocin ,STZ)诱导糖尿病大鼠病程 2wk时的血清 ,以DMEM培养基稀释 (5 0 ,10 0及 2 0 0mL·L-1)后作用于体外培养的大鼠RMCs,采用四唑盐 (tetrazolium ,MTT)还原比色法 ,测定MTT反应的吸光度 (A值 ) .结果 :大鼠血清能促进RMCs增殖 ,并呈现浓度依赖性 ,而早期糖尿病大鼠血清较对照组血清 (正常同龄大鼠的同浓度血清 )有显著的抑制增殖的作用 (P <0 .0 5 ) ,并随着血清浓度的增加 ,抑制率增加 ,5 0 ,10 0及 2 0 0mL·L-1时 ,其A值分别为 0 .2 0 8± 0 .0 2 0vs 0 .2 31± 0 .0 2 0 ,0 .2 6 7± 0 .0 13vs 0 .30 4± 0 .0 14和 0 .32 7± 0 .0 2 1vs 0 .380± 0 .0 2 4 ,其抑制率分别为 10 .0 % ,12 .2 %和 13.9% .结论 :早期糖尿病大鼠血清对RMCs的增殖有显著的抑制作用 .
