目的探讨广东汉族人群中脂联素基因(ADIPOQ)的单核苷酸多态性(SNP)与2型糖尿病的关联。方法采用SNPscanTM多重SNP分型试剂盒,对筛选的2个tagSNP(rs822396、rs3774261)在595名2型糖尿病患者和725名健康对照中进行基因分型,并应用χ2检验、Logistic回归模型等方法分析SNP位点与2型糖尿病的关联性。结果调整性别、BMI和甘油三酯等因素后,rs822396在等位基因关联分析和基因型关联分析中均无统计学意义(P>0.05),rs3774261在等位基因关联分析中亦无统计学意义[P=0.077,OR(95%CI)=1.18(0.98,1.41)],但在显性模型下的基因型关联分析中有统计学意义[P=0.028,OR(95%CI)=1.36(1.03,1.79)]。结论在广东汉族人群中,rs822396与2型糖尿病无关联,rs3774261与2型糖尿病的遗传易感性有关,但其具体机制尚需进一步探讨。
Objective To investigate the association of single nucleotide polymorphism (SNP) of adiponectin gene (ADIPOQ) with type 2 diabetes in Guangdong Han population. Methods The genotypes of 2 selected tagSNPs (rs822396, rs3774261) in 595 patients with type 2 diabetes mellitus and 725 healthy controls were genotyped by SNPscanTM multiple SNP typing kit and analyzed by χ2 test and Logistic regression model Association of SNP loci with type 2 diabetes mellitus. Results After adjusted for gender, BMI and triglyceride, rs822396 had no significant difference in allele association and genotype association (P> 0.05), rs3774261 was also not statistically significant in allele association analysis (P = 0.077, OR (95% CI) = 1.18 (0.98, 1.41)], but there was a statistically significant association in the dominant model (P = 0.028, OR 1.03, 1.79)]. Conclusion There is no association between rs822396 and type 2 diabetes mellitus in Guangdong Han population. Rs3774261 is associated with the genetic susceptibility to type 2 diabetes mellitus, but the exact mechanism needs to be further explored.
