以紫花补血草叶片为试验材料,采用不同的外植体消毒方法、不同激素配比的初代、继代和增殖培养基以及生根培养基,研究紫花补血草叶片的植株再生和快速繁殖方法。结果表明:紫花补血草的叶片外植体消毒以75%的酒精消毒10s加0.1%氯化汞浸泡8min为宜;叶组织脱分化成芽(初代培养)的适宜培养基为MS+BA 1.0mg/L+NAA 1.0mg/L;而芽的增殖和生根分别以MS+BA 0.1mg/L+NAA 0.1~0.5mg/L和1/2MS+NAA 0.1mg/L+蔗糖20g/L较优。通过叶片植株再生,建立了紫花补血草的组培快繁体系。
The method of regeneration and rapid propagation of Limonium sinense leaves was studied by using different methods of disinfection of explants, primary generation with different hormone ratio, subculture and multiplication medium and rooting medium. The results showed that it was appropriate to disinfect leaf explants of Limonium sinense by 75% alcohol disinfection for 10s and 0.1% mercuric chloride for 8 minutes. Suitable medium for leaf dedifferentiation to bud (primary culture) was MS + BA 1.0mg / L + NAA 1.0mg / L, while buds multiplication and rooting were better with MS + BA 0.1mg / L + NAA 0.1 ~ 0.5mg / L and 1 / 2MS + NAA 0.1mg / L + sucrose 20g / L, respectively. Through the regeneration of leaf plant, the tissue culture system of Limonium sinense was established.
