目的:观察芪箭消瘦汤对实验性自身免疫性甲状腺炎小鼠自身抗体、甲状腺病理、胀腔巨噬细胞诱生细胞因子的影响及作用机理。方法:运用人甲状腺球蛋白与完全弗氏佐剂混合免疫注射法模拟小鼠实验性自身免疫性甲状腺炎,用被动血凝法检测甲状腺自身抗体水平,收集腹腔巨噬细胞用放射免疫法测上清液肿瘤坏死因子和白细胞介素-6的含量,HE染色法观察小鼠早状腺组织病理改变。结果:芪箭消瘦汤明显抑制甲状腺自身抗体水平,减轻小鼠甲状腺组织淋巴细胞浸润。模型组小鼠腹腔巨嗜细胞诱生TNF、IL-6含量显著高于正常组(P<0.01)。芪箭小剂量组、雷公藤片组TNF显着低于模型组(P<0.01),大剂量组与模型组有差异(P<0.05)。芪箭大、小剂量组IL-6显著降低(P<0.01)。结论:芪箭消瘦汤可调节细胞因子的释放,改善桥本氏甲状腺炎的自身免疫紊乱,对小鼠实验性自身免疫性甲状腺炎疗效明显。
OBJECTIVE: To observe the effect and mechanism of Qijian Xiaoshen Decoction on autoantibodies, thyroid pathology, macrophage-induced cytokines in swelled cavities of experimental autoimmune thyroiditis in mice. METHODS: Experimental autoimmune thyroiditis in mice was immunized with human thyroglobulin and complete Freund’s adjuvant immunization. Autoimmune thyroid antibody levels were measured by passive hemagglutination method. Peritoneal macrophages were collected and measured by radioimmunoassay. The serum levels of tumor necrosis factor and interleukin-6 were measured by HE staining. RESULTS: Qijian Xiaoshen Tang significantly inhibited the level of thyroid autoantibodies and reduced lymphocyte infiltration in thyroid tissue of mice. The levels of TNF and IL-6 induced by mouse peritoneal macrophages in the model group were significantly higher than those in the normal group (P<0.01). The TNF of Xiaojian small dose group and Tripterygium wilfordii group was significantly lower than that of model group (P<0.01), and there was a difference between large dose group and model group (P<0.05). The IL-6 levels in the big and small dose groups were significantly decreased (P<0.01). CONCLUSIONS: Jianjianxiaoshen Decoction can regulate the release of cytokines, improve the autoimmune disorder of Hashimoto’s thyroiditis, and has obvious curative effect on experimental autoimmune thyroiditis in mice.
