为观察充血性心力衰竭 (CHF)患者外周血单核细胞 (PBMC)分泌肿瘤坏死因子α(TNF α)、白细胞介素 6 (IL 6 )和白细胞介素 1β(IL 1β)的水平以及卡托普利对其的影响 ,采用Ficoll Hypaque密度梯度离心法分离和纯化PBMC ,加入脂多糖和不同浓度的卡托普利 ,检测培养上清TNF α、IL 6和IL 1β。结果显示 ,CHF患者PBMC分泌TNF α、IL 6和IL 1β水平均较健康对照组显著升高 (P <0 0 5 ) ,并且随心功能分级的增加而增高 ;10 -3 mol/L及以上浓度的卡托普利可剂量依赖性地抑制PBMC分泌TNF α、IL 6和IL 1β(P <0 0 1)。提示CHF患者可能通过单核细胞分泌TNF α、IL 6和IL 1β的增加而影响心功能 ,而卡托普利治疗CHF的良好疗效 ,可能部分与其抑制细胞因子的作用有关
To investigate the levels of tumor necrosis factor α (TNFα), interleukin 6 (IL 6) and interleukin 1β (IL 1β) secreted by peripheral blood mononuclear cells (PBMCs) in patients with congestive heart failure (CHF) Puli on its impact, the use of Ficoll Hypaque density gradient centrifugation isolated and purified PBMC, adding lipopolysaccharide and different concentrations of captopril, culture supernatant TNFα, IL 6 and IL 1β. The results showed that the levels of TNFα, IL-6 and IL-1β secreted by PBMC in CHF patients were significantly higher than those in healthy controls (P <0.05), and increased with the increase of cardiac function classification. The levels of TNF-α, IL- Captopril inhibited PBMCs secretion of TNFα, IL-6 and ILβ in a dose-dependent manner (P <0.01). These results suggest that the CHF patients may affect the cardiac function by increasing the secretion of TNFα, IL-6 and ILβ by monocytes. The good curative effect of captopril in treating CHF may be partly related to the inhibition of cytokines
