目的探讨何首乌饮对运动性疲劳大鼠睾丸组织17β-羟基类固醇脱氢酶(17β-HSD)的影响。方法 50只雄性SD大鼠随机分为5组,A组为安静对照组;B组为安静何首乌饮组;C组为模型组;D组为何首乌饮治疗组;E组为何首乌饮预防组,每组10只。C、D和E组大鼠复制运动性疲劳动物模型,其中E组每天训练前给予何首乌饮20g/(kg.d)(含生药9.6kg/L)灌胃作为预防。模型成功后D组大鼠给予何首乌饮20g/(kg.d)(含生药9.6kg/L)灌胃治疗60d。放射免疫法检测各组大鼠血清睾酮浓度,分别采用免疫组织化学法、Western blotting和RT-PCR检测各组大鼠睾酮合成催化酶的表达变化。结果 17β-HSD蛋白的阳性产物主要表达在睾丸间质细胞。17β-HSD mRNA及蛋白表达变化:与C组相比,D组和E组17β-HSD有显著性提高(P<0.05),而D组和E组无差异(P>0.05)。结论何首乌饮可以提高睾酮合成限速酶17β-HSD的表达。
Objective To investigate the effects of Heshouwuyin on 17β-hydroxysteroid dehydrogenase (17β-HSD) in testis of rats with exercise-induced fatigue. Methods Fifty male Sprague-Dawley rats were randomly divided into 5 groups: group A was a quiet control group; group B was a quiet group containing polyphenols; group C was a model group; group D was a group treated by He Shouwu Yin; Each group of 10. Animals in groups C, D and E were used to replicate exercise-induced fatigue model. E group rats were given 20g / (kg.d) polygodial (9.6kg / L crude drug) before gavage. After successful model, rats in group D were given gavage for 20 d / d (containing crude drug 9.6 kg / L) for 60 days. Radioimmunoassay was used to detect the concentration of serum testosterone in each group. Immunohistochemistry, Western blotting and RT-PCR were used to detect the changes of the testosterone-producing enzymes in the rats. Results The positive product of 17β-HSD protein was mainly expressed in testicular stromal cells. 17β-HSD mRNA and protein expression: Compared with group C, 17β-HSD in group D and group E increased significantly (P <0.05), but there was no difference between group D and group E (P> 0.05). Conclusion Polygonum multiflorum Thunb can increase the expression of 17β-HSD, a rate-limiting enzyme in testosterone synthesis.
