目的探讨不同室温下EDTA修复时间对P504S蛋白在前列腺癌组织中表达的影响情况。方法分析30例前列腺癌患者临床资料,将病理标本分别在3种不同室温下进行抗原修复,分别每例标本切9张片。观察不同室温下进行抗原修复显色反应和背景显色情况。结果室温8~10℃抗原修复30 min,室温18~20℃抗原修复20、25 min显色反应的准确率最高,背景显色效果最好,P<0.05,差异均有统计学意义。结论室温8~10℃抗原修复30 min,18~20℃抗原修复20、25 min对P504S蛋白在前列腺癌组织中表达显色效果最好,为病理诊断提供准确依据,值得临床推广应用。
Objective To investigate the effect of different repair time of EDTA at room temperature on the expression of P504S protein in prostate cancer. Methods The clinical data of 30 patients with prostate cancer were analyzed. The pathological specimens were subjected to antigen retrieval at 3 different room temperatures. Nine specimens were cut in each specimen. Observed at different room temperature for antigen repair color reaction and background color. Results After the antigen was fixed at 8-10 ℃ for 30 min at room temperature, the color reaction was the highest at 20 and 25 min after antigen retrieval at room temperature (18-20 ℃), and the best background color was obtained. The difference was statistically significant (P <0.05). Conclusion Antigen repair at room temperature of 8 ~ 10 ℃ for 30 min and antigen reversion at 18 ~ 20 ℃ for 20 and 25 min have the best effect on the expression of P504S protein in prostate cancer tissues, which provides an accurate basis for pathological diagnosis and is worthy of clinical application.
