以夏天无块茎为试材,采用不同浓度秋水仙素(0.00%、0.05%、0.10%、0.20%)添加2%二甲基亚砜(DMSO)分别浸泡24、48、72h的方法,研究了不同浓度秋水仙素和不同处理时间对染色体加倍的影响。结果表明:以0.20%秋水仙素处理24h较优,成活率为31.4%,加倍率为50.0%,其次为0.10%秋水仙素处理24、48h和0.20%秋水仙素处理48h。其中,未加倍的植株染色体数为2n=28,加倍后的植株为2n=56。
The tuberous tuber of summer was used as experimental material to study the effect of different concentrations of colchicine (0.00%, 0.05%, 0.10%, 0.20%) on the addition of 2% dimethylsulfoxide (DMSO) for 24, 48 and 72h respectively. Effect of Different Concentrations of Colchicine and Different Processing Times on Chromosome Doubling. The results showed that the treatment with 0.20% colchicine for 24 hours was superior with the survival rate of 31.4% and the doubling rate of 50.0%, followed by 0.10% colchicine treatment for 24,48 hours and 0.20% colchicine treatment for 48 hours. Among them, the number of non-doubling plant chromosome 2n = 28, doubling the plant 2n = 56.
