小鼠皮下移植性肝癌模型的热消融研究

目的:探讨建立适合微波消融研究的肝癌小鼠模型。方法:将C57BL/6J小鼠随机分为4组(n=10),分别皮下注射不同数量(5×105,1×106,2×106,5×106)的Hepa1-6细胞,比较各组成瘤率;选择成瘤率理想的细胞数再次造模,待瘤体长径达8~10 mm时将模型小鼠随机分为4组(n=10),对瘤体行微波热消融处理,瘤体边缘温度分别控制在60℃,55℃,50℃和45℃,消融时间为3 min,比较各组完全消融率及消融前后的瘤体病理变化。结果:2×106的Hepa1-6细胞皮下注射组成瘤率高且适合消融操作;60℃消融组出现组织碳化,其余各组比较,55℃消融组完全消融率最高,复发率最低(P<0.01)。结论:对小鼠Hepa1-6细胞皮下移植性肝癌模型行55℃×3 min的微波热消融有效、可行。 Objective: To establish a mouse model of hepatocellular carcinoma suitable for microwave ablation. Methods: C57BL / 6J mice were randomly divided into 4 groups (n = 10). Hepa1-6 cells of different numbers (5 × 105, 1 × 106, 2 × 106, 5 × 106) The rate of tumor was selected. The number of cells with ideal tumor formation rate was re-modeled. When the length of tumor reached 8-10 mm, the model mice were randomly divided into 4 groups (n = 10) The edge temperature of the tumor was controlled at 60 ℃, 55 ℃, 50 ℃ and 45 ℃ respectively, and the ablation time was 3 min. The complete ablation rate and pathological changes of the tumor before and after ablation were compared. Results: The subcutaneous injection of 2 × 106 Hepa1-6 cells resulted in a high rate of tumorigenicity and suitable for ablation. The tissue carbonization occurred in 60 ℃ ablation group, and the complete ablation rate was the lowest in 55 ℃ ablation group (P <0.01) ). Conclusion: It is feasible and feasible to microwave heat ablate Hepa1-6 mouse subcutaneous transplanted hepatocarcinoma model at 55 ℃ for 3 min.
