目的:探讨中药对延缓DHE耐受小鼠脑内的下丘脑以及杏仁核CCKm RNA 表达的影响。方法:采用热板法,以添足底反应潜伏期为痛阈。Q500IW 软包分析小鼠脑内CCKm RNA 表达量。结果:小鼠接受混合中药后,从第3d 开始到第7d 为止,AUC值一直保持较高的水平。第7d AUC值与单纯DHE组比较,前者为(4458.4±1357.6),而后者(1773.6±1144.2),两者存在差异有显著意义(P< 0.05)。CCKm RNA 原位杂交信号经图像处理结果:下丘脑和杏仁核CCKm RNA 表达,耐受组分别为103.92和181.19,明显高于混合中药组和对照组(45.72和70.13)P< 0.05,而中药组为51.03和92.29与对照组相比无明显差异P> 0.05。结论:DHE耐受小鼠,脑内下丘脑和杏仁核CCKm RNA 表达的增加,可能是产生DHE耐受的原因之一。
OBJECTIVE: To investigate the effect of traditional Chinese medicine (TCM) on delaying the expression of CCK mRNA in hypothalamus and amygdala of DHE-tolerant mice. Methods: The hot plate method was used to add the plantar reaction latency as the pain threshold. The Q500IW soft package analyzes the expression of CCKm RNA in the mouse brain. RESULTS: After the mice received the mixed herbs, the AUC value remained high from the 3rd day to the 7th day. The 7th day of AUC was compared with that of DHE alone. The former was (4458.4±1357.6), while the latter was (1773.6±1144.2). There was a significant difference between the two groups (P<0.05). Image processing results of CCKm RNA in situ hybridization signal: CCKm RNA expression in hypothalamus and amygdala was 103.92 and 181.19 in the tolerant group, which was significantly higher than that of the mixed traditional Chinese medicine group and the control group (45.72 and 70.13). ) P < 0.05, while the Chinese medicine group 51.03 and 92.29 compared with the control group no significant difference P> 0.05. CONCLUSIONS: Increased expression of CCK mRNA in hypothalamus and amygdala of DHE-tolerant mice may be one of the causes of DHE tolerance.
