上皮细胞间叶化(epithelial-mesenchymal transition,EMT)与肿瘤侵袭转移密切相关,转化生长因子-β(transforming growth factor-beta,TGF-β)已被证实为肿瘤EMT的主要诱导剂。该研究旨在探讨TGF-β1对人前列腺癌PC3细胞EMT发生的诱导作用。经TGF-β1处理的PC3细胞,在相差倒置显微镜下观察到细胞形态由上皮型向间叶型转化;细胞划痕和Transwell实验检测细胞的迁移能力较对照组显著增强;Real-time PCR、Western blot和细胞免疫荧光验证TGF-β1能上调间叶型标记蛋白的表达及下调上皮型标志蛋白的表达。进一步发现,TGF-β1对转录因子Snail的表达没有明显的影响,但是能促进Snail的入核,干扰Snail能逆转TGF-β1对PC3细胞EMT发生的诱导作用。该研究表明,TGF-β1能够介导Snail的入核而激活Snail,促进前列腺癌PC3细胞的发生。
Epithelial-mesenchymal transition (EMT) is closely related to tumor invasion and metastasis. Transforming growth factor-beta (TGF-beta) has been identified as the main inducer of EMT in tumors. The aim of this study was to investigate the induction of TGF-β1 on EMT in human prostate cancer PC3 cells. Transforming cells from epithelial to mesenchymal cells transformed by TGF-β1 treated PC3 cells were observed by inverted phase contrast microscope. Cell migration and cell migration were significantly enhanced by cell scratch assay and Transwell assay. Real-time PCR, Western The results of Western blot and immunofluorescence showed that TGF-β1 upregulated the expression of mesenchymal marker protein and down-regulated the expression of epithelial marker protein. It was further found that TGF-β1 had no significant effect on the expression of Snail, but could promote the entry of Snail and the interference of Snail on the induction of TGF-β1 on the induction of EMT in PC3 cells. The study shows that, TGF-β1 can lead Snail into the nucleus and activate Snail, promote prostate cancer PC3 cells.
