利用免疫组织化学技术对黄瓜离休茎段嫁接体内吲哚乙酸(IAA)的动态变化进行定位研究.结果表明冰冻干燥-气体固定法较溶液化学固定法固定IAA的效率高.培养基中不含激素时,嫁接体组织中银颗粒标记较少,嫁接体发育过程中IAA的动态变化不明显.培养基中合适当激素时,银颗粒标记较多,其数量的变化与嫁接体发育密切相关.对激素定位研究方法及IAA在嫁接体发育过程中的作用进行了讨论.
The dynamic changes of indole acetic acid (IAA) in the grafted cucumber leaves were studied by immunohistochemistry. The results show that the lyophilization - gas fixation method is more efficient than the solution chemical fixation method for IAA fixation. In the absence of hormones in the medium, the silver particles in the grafted tissue were less labeled, and the dynamic changes of IAA during the development of the grafted body were not obvious. When the appropriate hormone in the medium, silver particles more markers, the number of changes closely related to the development of grafts. The methods of hormone localization and the role of IAA in the development of grafts were discussed.
