目的:研究济脉通与硝苯地平合用对SHR血压的影响,并探讨其降压作用机制,阐述其联合用药的特点或优势。方法:采用自发性高血压大鼠(SHR)模型,设立模型组、济脉通组(450 mg.kg-1)、硝苯地平组(5 mg.kg-1)、济脉通与硝苯地平联合用药组(300 mg.kg-1+0.5 mg.kg-1),连续给药8周后,测定血压、心率、氧化、血脂及激素指标。结果:与模型组比较,3个给药组均有降低SHR收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP)的作用;硝苯地平组使心率(HR)加快(P<0.05),济脉通组和联合用药组对HR则无显著性影响。联合用药组明显降低尿素(URE)、丙二醛(MDA)、血浆内皮素(ET)及血浆血管紧张素II(AngⅡ)含量(P<0.05,P<0.05,P<0.05,P<0.01),明显提高一氧化氮(NO)、环磷酸腺苷(CAMP)含量和超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)含量(P<0.05),是其可能的降血压机制。结论:济脉通与硝苯地平联合用药对自发性高血压大鼠有明显的协同降压作用,研究结果表明中西医联合用药对高血压病的治疗具有优势。
Objective: To study the effect of combination of Jimai Tong and nifedipine on blood pressure of SHR and to explore its mechanism of antihypertensive effect, and to explain the characteristics or advantages of its combination therapy. Methods: The spontaneous hypertensive rats (SHR) model was established. The model group was established with JiMiTong group (450 mg.kg-1), nifedipine group (5 mg.kg-1) (300 mg.kg-1 + 0.5 mg.kg-1). After 8 weeks of continuous administration, blood pressure, heart rate, oxygenation, blood lipid and hormones were measured. Results: Compared with the model group, the SBP, DBP and MAP decreased in all three groups. The nifedipine group increased the heart rate (HR) (P < 0.05). The Jimai Tong group and combination group had no significant effect on HR. The combined treatment group significantly reduced the content of URE, MDA, ET and AngⅡ (P <0.05, P <0.05, P <0.05, P <0.01) , Significantly increased nitric oxide (NO), cyclic adenosine monophosphate (CAMP) and superoxide dismutase (SOD), nitric oxide synthase (NOS) content . Conclusion: The combination of Jimantong and nifedipine has obvious synergistic antihypertensive effects on spontaneously hypertensive rats. The results show that the combination of Chinese and Western medicine has the advantage on the treatment of hypertension.
