目的:研究治偏痛对硝酸甘油(NTG)诱导的偏头痛模型小鼠搔头次数和血清中一氧化氮(NO)、降钙素基因相关肽(CGRP)、多巴胺(DA)及5-羟色胺(5-HT)含量的影响。方法:治偏痛分别以0.4、0.2、0.1g生药/kg的剂量(即分别为高、中、低组)鼻腔给药3天,末次给药后皮下注射硝酸甘油建立偏头痛小鼠模型,空白对照组和模型组鼻腔给予同体积蒸馏水,记录小鼠前肢搔头的次数及测定血清中NO、CGRP、DA、5-HT。结果:与模型对照相比,治偏痛高、中、低剂量均可显著减少偏头痛模型小鼠前肢搔头次数(P<0.05,P<0.01,P<0.001),同时高剂量显著降低血清中NO、CGRP、5-HT、DA含量(P<0.05,P<0.01),中剂量显著降低血清中NO、CGRP、5-HT、DA含量(P<0.05),低剂量显著降低血清中DA含量(P<0.05),起效剂量为0.1g生药/kg,起效范围为0.1-0.4g生药/kg。结论:治偏痛可有效预防硝酸甘油诱导的偏头痛,其机制可能是通过下调血清中NO、CGRP、5-HT和DA的水平,改善了血管舒缩功能障碍,缓解偏头痛的发生。
Objective: To study the effect of treatment of migraine on the number of scratching and serum NO, CGRP, DA and serotonin in mice with migraine induced by nitroglycerin (NTG) (5-HT) content of the impact. Methods: The patients were treated by intranasal administration of 0.4,0.2 and 0.1g crude drug / kg (ie, high, medium and low groups respectively) for 3 days. After the last administration, nitroglycerin was injected subcutaneously to establish a mouse model of migraine. The same volume of distilled water was administered to the nasal cavity of the blank control group and the model group. The frequency of scratching the forelimbs of the mice was recorded and the levels of NO, CGRP, DA and 5-HT in the serum were measured. Results: Compared with the model control, the treatment of high, medium and low dose of migraine pain can significantly reduce the frequency of head forelimb scratching in migraine model mice (P <0.05, P <0.01, P <0.001) The contents of NO, CGRP, 5-HT and DA in serum were significantly decreased (P <0.05, P <0.01) and the levels of NO, CGRP, 5-HT and DA in serum were significantly decreased (P <0.05), the effective dosage was 0.1g crude drug / kg, the effective range was 0.1-0.4g crude drug / kg. Conclusion: The treatment of migraine can effectively prevent the migraine induced by nitroglycerin. The mechanism may be that it can ameliorate the vasomotor dysfunction and relieve the migraine by reducing the levels of NO, CGRP, 5-HT and DA in the serum.
