目的:研究透明质酸(HA)对CNE-2Z裸鼠移植瘤血管内皮细胞生长因子(VEGF)和其受体(flk-1)表达的影响。方法:将CNE-2Z癌细胞接种到裸鼠皮下,并于原位注射透明质酸(HA)(每次300μg,共1800μg),每周测瘤直径,5周后处死动物并称瘤重。取移植瘤作冰冻切片,并进行VEGF及flk-1免疫组化染色。结果:癌细胞胞浆内VEGF阳性,癌细胞坏死区阳性,癌间质和癌浸润周边处毛细血管内皮细胞阳性,但对照组和实验组VEGF表达未见明显差异;实验组和对照组癌组织内及癌浸润周边处毛细血管内皮细胞flk-1阳性,但数目及分布不同;实验组第一周瘤体积明显增大。结论:HA对癌细胞表达VEGF无明显影响,但影响毛细血管的flk-1的表达及血管形成,HA能促进移植瘤的早期生长。
Objective: To investigate the effect of hyaluronic acid (HA) on the expression of vascular endothelial growth factor (VEGF) and its receptor (flk-1) in CNE-2Z xenografted nude mice. Methods: CNE-2Z cancer cells were inoculated subcutaneously in nude mice and in situ were injected with hyaluronic acid (HA) (300μg each) for a total of 1800μg. The diameter of the tumor was measured weekly. After 5 weeks, the animals were sacrificed and weighed. Tumor xenografts were frozen sectioned and immunohistochemically stained for VEGF and flk-1. Results: The positive cells in the cytoplasm of the cancer cells were positive for VEGF, the positive cells in the necrotic area of the cancer cells, the positive expression of the capillary endothelial cells in the periphery of the cancer cells and the cancer infiltration. However, there was no significant difference in VEGF expression between the control group and the experimental group. The number and distribution of flk-1 in capillary endothelial cells around and around the cancer infiltration were different. The tumor volume of the experimental group increased significantly in the first week. CONCLUSION: HA has no obvious effect on the expression of VEGF in cancer cells, but affects the expression of flk-1 and angiogenesis in capillary vessels. HA can promote the early growth of xenografts.
