目的:用细胞学方法评价左金丸、反左金丸、茱萸丸、甘露散、交泰丸等中药复方的寒热药性。方法:用噻唑蓝(MTT)比色法考察5种中药复方对He La细胞和SGC-7901细胞体外增殖的影响,倒置显微镜观察其对细胞形态特征的影响。结果:反左金丸、茱萸丸在所选质量浓度5-800μg/m L范围内对He La细胞和SGC-7901细胞的生长增殖表现出促进作用,表现为热性;左金丸、甘露散、交泰丸在所选质量浓度5-800μg/m L范围内对He La细胞和SGC-7901细胞的生长增殖表现出抑制作用,随着浓度的增大,抑制率逐渐增大,表现为寒性。形态学观察表明,左金丸、甘露散与交泰丸能使细胞密度减小,固缩变圆;反左金丸、茱萸丸可使细胞密度增加,结构致密,生长旺盛。台盼蓝染色表明,5种中药复方对这两种细胞没有细胞毒作用。结论:5种中药复方的寒热药性细胞学方法评价结果与文献记载一致,说明细胞学方法能够用于中药复方寒热药性的评价,为研究中药复方寒热药性提供了实验依据和方法学参考。
OBJECTIVE: To evaluate the cold-heat medicinal properties of Zuojin pills, Zuozuo jin pills, Zhuyu pills, Ganluosan, Jiaotai pills and other traditional Chinese medicine compounds by cytology. Methods: MTT assay was used to investigate the effects of five kinds of traditional Chinese medicine on proliferation of He La cells and SGC-7901 cells in vitro. The morphological changes of cells were observed by inverted microscope. RESULTS: Anti-Zuogan pill and Fructus corni were found to promote the growth and proliferation of HeLa cells and SGC-7901 cells in the range of 5-800 μg / mL, Tarim inhibited the growth of HeLa cells and SGC-7901 cells in the concentration range of 5-800 μg / mL, and the inhibitory rate increased gradually with the increase of concentration. Morphological observation showed that Zuojinwan, Ganluosan and JiaoTaiwan can reduce the cell density and shrinkage; the anti-Zuojin pill and Fructus corni could increase the cell density, the dense structure and the strong growth. Trypan blue staining showed that five kinds of traditional Chinese medicine compound have no cytotoxic effect on these two kinds of cells. CONCLUSION: The results of cold and heat cytotoxicity assay of 5 kinds of Chinese traditional medicine compounds are consistent with the literature records, indicating that cytological methods can be used to evaluate the cold-heat medicinal properties of TCM compound prescriptions, providing experimental basis and methodological reference for studying the cold-hot medicinal properties of TCM compound prescriptions.
