用~(125)Ⅰ—乳过氧化物酶法标记恶件疟原虫裂殖体(子),TritonX—100提取疟原虫表面标记抗原。抗恶性疟单克隆抗体9_4D_1与上述恶性症原虫膜抗原提取物作免疫沉淀分析,并用单克隆抗体9_4D_1制备免疫吸附剂,通过亲和层析自恶性疟原虫提取物中分离纯化相应的靶抗原。结果表明,免疫沉淀的靶抗原分子量为55kd,48kd和25kd的多肽,与亲和层析纯化出的靶抗原基本一致.
Plasmodium falciparum Schizonts (Triton X-100) were labeled with ~ (125) I-lactoperoxidase and Plasmodium malariae surface antigen was extracted by Triton X-100. Anti-Plasmodium falciparum monoclonal antibody 9_4D_1 was immunoprecipitated with the above protozoal protozoal membrane antigen extract, and the monoclonal antibody 9_4D_1 was used to prepare the immunosorbent. The corresponding target antigen was isolated and purified from the Plasmodium falciparum extract by affinity chromatography. The results showed that the immunoprecipitated target antigen molecular weight of 55kd, 48kd and 25kd peptide, and affinity chromatography purified target antigen is basically the same.
