c-Jun二聚化蛋白(JDP2)是AP-1家族成员,参与基因的转录抑制.为探讨JDP2在神经母细胞瘤分化过程中的作用,构建了稳定整合有JDP2全长编码区的SH-SY5Y细胞株(SY5Y-JDP2).实验结果显示,JDP2能促进SH-SY5Y细胞的分化.与对照细胞相比,SY5Y-JDP2细胞在S期比例降低,生长速率迟缓,并且表现p21高水平表达,在分化过程中cyclin D1降低.由此可见,JDP2对生长的抑制作用可能是SH-SY5Y细胞撤出细胞周期进入分化的主要原因.
c-Jun dimerization protein (JDP2) is a member of the AP-1 family and is involved in the transcriptional repression of genes.In order to investigate the role of JDP2 in the differentiation of neuroblastoma, a JDP2 cell line stably integrated with the JDP2 full- SY5Y cell line SY5Y-JDP2.The results showed that JDP2 can promote the differentiation of SH-SY5Y cells.Compared with the control cells, the SY5Y-JDP2 cells decreased in S phase and the growth rate was slow, and showed high expression of p21, In the process of differentiation, the decrease of cyclin D1 was observed, which indicated that the inhibitory effect of JDP2 on the growth of SH-SY5Y cells may be the main reason for the differentiation of SH-SY5Y cells into the cell cycle.
