Ｓｐｒａｇｕｅ－Ｄａｕｌｃｙ大鼠３０只，随机分成①中毒剂量组１３只，②治疗剂量组，③对照组８只。分别皮下注射庆大霉素（ＧＭ）：①组４０ｍｇ／ｋｇ，②组５ｍｇ／ｋｇ，③组生理盐水等体积，观察用药前后血、尿标本测定尿磷酸二酯酶Ⅰ（ＰＤＥⅠ）、Ｎ－乙酰－β－氨基葡萄糖苷酶（ＮＡＧ），碱性磷酸酶（ＡＬＰ），β２－微球蛋白（β２Ｍ）的变化及大鼠肾组织病理学改变。结果：①组从第４ｄ开始，３种尿酶均显著性增高（Ｐ＜０．０１），持续至停药后１２ｄ～１８ｄ；②组尿ＮＡＧ、ＰＤＥＩ分别在４ｄ、６ｄ开始升高（Ｐ＜０．０１）至停药后８ｄ、１４ｄ，恢复治疗前水平，ＡＬＰ增加缓慢，仅在停药前才明显升高，并很快降低。病理检查结果：①组肾小球充血，部分近曲小管上皮细胞坏死；②组病变较轻，对照未见明显病变。提示：尿ＰＤＥＩ用于反应药物ＧＭ造成急性肾损伤较ＮＡＧ、ＡＬＰ更敏感和稳定。
Thirty Sprague-Daulcy rats were randomly divided into ① poisoning dose group of 13, ② therapeutic dose group, ③ control group of 8 rats. Gemcitabine (GM) was injected subcutaneously into the rats in each group: ①40mg / kg group, ②5mg / kg group, ③groups of equal volume of normal saline. The urinary phosphodiesterase Ⅰ (PDEⅠ) Acetyl-β-glucosaminidase (NAG), alkaline phosphatase (ALP), β2-microglobulin (β2M) changes and rat renal tissue pathology. Results: ①The urinary enzymes of three groups increased significantly (P <0.01) from the 4th day to the 12th ~ 18th day after stopping the treatment. ② The levels of NAG and PDEI increased at the 4th and 6th day (P <0.01) to the 8d and 14d after stopping the treatment, the levels of ALP before and after treatment were restored. The ALP increased slowly and only increased significantly before stopping, and soon decreased. Pathological examination results: ① group of glomerular hyperemia, part of the proximal tubule epithelial cell necrosis; ② group lesion was lighter, no obvious pathological changes. Hint: Urinary PDEI is more sensitive and stable than NAG and ALP in acute renal injury induced by GM.
