应用基于IIB型限制性内切酶的RAD(2b-RAD)技术,对‘沪五彩花糯1号’及其亲本材料进行了简并基因组测序。利用RADtyping分型策略,通过与‘B73’参考基因组比对,我们在3个玉米样品的共同测序标签内筛选获得了9 726个单核苷酸多态性(SNP)位点。在此基础上,利用9 707个位于玉米染色体上的SNP位点对‘沪五彩花糯1号’及其亲本进行了基因型分析,开发出可用于‘沪五彩花糯1号’品系特异性鉴定的9个SNP分子标记。这些研究结果为‘沪五彩花糯1号’品系特异性鉴定和优质性状遗传组成的分子机理研究奠定了基础。
Degenerate genomic sequencing was performed on ’Shanghai Wucaihuanuo 1’ and its parental material using the type IIB restriction endonuclease RAD (2b-RAD) technique. Using the RADtyping typing strategy, we screened 9 726 single nucleotide polymorphisms (SNPs) within a co-sequencing tag of 3 maize samples by aligning with the ’B73’ reference genome. On this basis, 9 707 SNPs locating on maize chromosome were used to analyze the genotypes of ’Shanghai Wucaihuanuo 1’ and its parents, Nine SNP markers identified. These results laid the foundation for the molecular characterization of ’Shanghai Wucaihuanuo 1’ line and the molecular mechanism of the genetic composition of high quality traits.
