参芪泄浊饮对大鼠腺嘌呤肾间质纤维化模型大鼠肾组织MMP-1、TIMP-1基因表达影响随机平行对照研究

[目的]研究参芪泄浊饮对腺嘌呤致大鼠慢性肾衰肾组织基质金属蛋白酶1(MMP-1)、基质金属蛋白酶抑制剂1(TIMP-1)基因表达的影响。[方法]SPF级雄性SD大鼠54只,采用Yokozawa法复制慢性肾衰大鼠模型,随机分为参芪泄浊饮高剂量组、参芪泄浊饮常规剂量组、氯沙坦组、模型组,分别予高剂量组、常规剂量组、氯沙坦组以参芪泄浊饮水煎液20m L/kg·d、10m L/kg·d、氯沙坦组9mg/kg·d,模型组与空白组大鼠均予纯净水10m L/kg·d,连续灌胃干预8周,ELISA法测量血肌酐(Scr)、尿素氮(BUN)、丙氨酸转氨酶(ALT);荧光定量PCR方法检测肾组织中MMP-1、TIMP-1。[结果]1Scr、BUN各组大鼠较空白组均有升高(P<0.05),氯沙坦组明显低于模型组(P<0.05);Scr高剂量组大鼠明显低于常规剂量组、氯沙坦组及模型组大鼠(P<0.05),常规剂量组与氯沙坦组亦低于模型组(P<0.05);BUN变化趋势与Scr相同,高剂量组降低不明显(P>0.05)。2MMP-1 m RNA各组均高于空白组(P<0.05),高剂量组显著高于常规剂量组、氯沙坦组及模型组(P<0.05),常规剂量组与氯沙坦组也显著高于模型组(P<0.05);TIMP-1 m RNA各组均高于空白组(P<0.05),高剂量组明显低于常规剂量组、氯沙坦组及模型组(P<0.05),常规剂量组与氯沙坦组明显低于模型组(P<0.05)。[结论]参芪泄浊饮可通过促进MMP-1 m RNA及抑制TIMP-1 m RNA表达,使细胞外基质(extracellular matrix,ECM)的降解增加,堆积减少,进而减缓CRF的进一步发展,保护肾脏。 [Objective] To investigate the effect of Shenqi Xiezhuo Yin on gene expression of matrix metalloproteinase 1 (MMP-1) and matrix metalloproteinase 1 (TIMP-1) in rats with chronic renal failure induced by adenine. The change tendency of BUN was the same as that of Scr, and the decrease was not obvious in high dose group > 0.05). The levels of 2MMP-1 m RNA in each group were significantly higher than those in the blank group (P <0.05). The high-dose group was significantly higher than the conventional dose group, the losartan group and the model group (P <0.05) (P <0.05). The levels of TIMP-1 mRNA in each group were significantly higher than those in the blank group (P <0.05), and those in the high dose group were significantly lower than those in the normal dose group, losartan group and model group ), The conventional dose group and losartan group were significantly lower than the model group (P <0.05). [Conclusion] Shenqi Xiezhuo Yin can increase the degradation of extracellular matrix (ECM) and decrease the accumulation of MMP-1 mRNA and TIMP-1 mRNA, thereby further reducing the progression of CRF and protecting kidney.
