目的:探讨应用简便、快速及无害化的非核素银染端粒重复序列扩增法(TRAP)检测人细胞端粒酶活性。方法:与核素TRAP相比较,采用非核素银染TRAP检测了293细胞,并检测了经RNase和加热处理的阴性对照标本和QGY7701、SMMC-77212株人肝癌细胞。结果:10个以上的293细胞均为阳性,而RNase和加热处理的标本均阴性;2株人肝癌细胞均阳性;非核素银染TRAP比核素TRAP快10h左右。结论:非核素银染的TRAP是特异、敏感及快速的端粒酶活性检测方法,证实2株人肝癌细胞株均有端粒酶活性表达。
OBJECTIVE: To investigate the telomerase activity in human cells using a simple, rapid, and destructive method using non-nuclear silver stained telomere repeat amplification (TRAP). METHODS: 293 cells were stained with non-nuclear silver-stained TRAP compared to radionuclide TRAP, and negative control samples treated with RNase and heat and QGY7701, SMMC-77212 human hepatoma cells were detected. RESULTS: More than 10 293 cells were positive, but RNase and heat-treated specimens were negative; 2 human hepatoma cells were positive; non-nuclear silver-stained TRAP was about 10 h faster than radionuclide TRAP. Conclusion: The non-nuclear silver-stained TRAP is a specific, sensitive and rapid method for detecting telomerase activity, confirming that the expression of telomerase activity in both human hepatoma cell lines is confirmed.
