目的总结本实验室SPF级环境中NC系裸小鼠繁殖和应用情况。方法用♀Nu/+与♂Nu/Nu交配法繁殖仔鼠后随机观察10窝裸小鼠的繁殖情况,然后用人脑胶质瘤组织或细胞悬液在裸小鼠体内建立原位和异位移植瘤模型,并分析模型生物学特性及实验治疗效果。结果 NC系裸小鼠每胎平均产仔9.1只,Nu/Nu占1/2,总离乳率为94.5%,年存活率达100%,未检测到病菌和寄生虫等。原位移植瘤能够复制其原发人脑胶质瘤侵袭性的生长特性,治疗后可以明显看到移植瘤缩小及血管生成抑制。结论 SPF级环境中用♀Nu/+与♂Nu/Nu交配法能稳定繁殖裸小鼠;裸小鼠为人脑恶性肿瘤模型的建立及实验治疗提供了理想的载体。
Objective To summarize the breeding and application of NC nude mice in SPF environment in our laboratory. Methods The reproduction of offspring of 10 nude mice was observed after breeding offspring by ♀Nu / + mating with ♂Nu / Nu. Then the nude mice were inoculated with human brain glioma tissue or cell suspension in situ and ectopic Xenograft model and analyze the model biological characteristics and experimental treatment effect. Results The NC nude mice each had an average of 9.1 litters per litter. Nu / Nu accounted for 1/2 of the total. The total rate of maternal milk was 94.5%. The annual survival rate was 100%. No pathogens, parasites and so on were detected. In situ xenografts replicate the aggressive growth characteristics of their primary human gliomas, and the reduction of transplanted tumors and angiogenesis inhibition are evident after treatment. Conclusion Nude mice can be propagated successfully by mating ♀Nu / + and ♂Nu / Nu in SPF environment. Nude mice provide an ideal carrier for the establishment and experimental treatment of human brain malignant tumor model.
