通过RACE技术从中间锦鸡儿中克隆得到一个WRKY转录因子编码基因CiWRKY2,该基因gDNA为3 224 bp,含有4个内含子;cDNA长2 032 bp,开放阅读框1 725 bp,可以编码574个氨基酸,蛋白分子量大小约为63.29 kD,等电点为7.65.系统进化分析发现,CiWRKY2与豆科植物WRKY相似度最高,属于第一类WRKY蛋白.亚细胞定位显示CiWRKY2蛋白定位于细胞核.实时荧光定量结果表明CiWRKY2主要在叶、根中表达,并且该基因转录水平的表达受到干旱、盐碱、高温、低温和ABA诱导,表明CiWRKY2基因可能在植物抗逆中起到一定作用.“,”A WRKY encoding gene was cloned by RACE technique from Caragana intermedia,designated as CiWRKY2.The gDNA of CiWRKY2 was 3 224 bp,containing four introns;the cDNA was 2 032 bp including a 1 726 bp open reading frame,which encoded a 574-amino acid protein with an estimated molecular mass of 63.29 kD and an isoelectric point of 7.65.Phylogenetic analysis showed that CiWRKY2 shared high identity with the WRKY proteins from leguminous plants and belonged to group Ⅰ of the WRKY transcription factor family.CiWRKY2 localized in the nuclei and mainly expressed in leaves and roots.The expression patter analyzed by qRT-PCR showed that CiWRKY2 transcript was induced under drought,NaCl,high pH,cold,heat and ABA treatments.It suggested that CiWRKY2 might play a role in plant stress tolerance.
