以蕙兰×台兰种间杂交的种子为材料,研究适合于种子无菌播种、原球茎分化芽、生根壮苗的不同培养基.结果表明:在5种用于无菌播种的培养基中,添加IAA10mg/L的C1培养基,种子萌动率达3%,原球茎由培养120天的10.13个/瓶,平均直径1.63mm到220天增至19.86个/瓶,平均直径4.30mm;BA1~2mg/L有利于原球茎分化芽,分化率达100%以上;NAA2mg/L使生根壮苗率达100%.添加活性炭1.5~3g/L对种子成苗过程效果较好,在芽分化与生根壮苗培养中,减少MS大量元素的使用量有利于生长.兰花试管苗移栽成活率为80%,已获得4~5年生的杂种苗.
The seeds of Cymbidium × Tailan interspecific hybrids were used as materials to study the different media suitable for seed sowing, protocorm bud differentiation and rooting. The results showed that the seed germination rate was up to 3% with 10 mg / L IAA in medium of 5 kinds of asepsis for sowing, and the diameter of protocorm was 10.13 / 63mm to 220 days increased to 19.86 / bottle, the average diameter of 4.30mm; BA1 ~ 2mg / L is conducive to the formation of protocorm bud differentiation rate of 100% or more; NAA2mg / L so that the strong rooting rate of 100%. Adding 1.5 ~ 3g / L activated charcoal had a better effect on the seedling formation process. Reducing the amount of MS elements in the bud differentiation and rooting and seedling culture was favorable to the growth. Orchid seedling transplanting survival rate of 80%, has been 4 to 5 years of hybrid seedlings.
