目的建立一种稳定高效获得鹿茸片DNA基因组的提取方法,为相似形态中药总DNA提取奠定基础,便于应用分子生物学进行药材鉴定。方法将乌鲁木齐市售鹿茸饮片选取3份为材料,通过两种试剂盒和CTAB三种不同方法提取,提取的DNA再进行琼脂糖凝胶电泳法、紫外分光光度法进行浓度和纯度检测。结果 3种方法均可以从市售鹿茸饮片中提取到基因组DNA,但天根组织提取试剂盒得到的基因组DNA的纯度和浓度最高,提取效果最佳;CTAB法提取效果相对最差。结论天根组织提取试剂盒基因组DNA提取方法操作简便、稳定、高效,提取的DNA效果较好,纯度和浓度检查均合格。
OBJECTIVE To establish a stable and efficient extraction method for DNA genome of pilose antler pieces and lay a foundation for the extraction of total DNA from similar forms of traditional Chinese medicine and facilitate the identification of medicinal materials by molecular biology. Methods Three samples of commercially available deer horn pieces were collected from Urumqi, and were extracted by two kinds of kits and CTAB. The DNA was extracted by agarose gel electrophoresis and UV spectrophotometry. Results All three methods could extract genomic DNA from commercially available antler pieces. However, the genomic DNA obtained from the root extraction kit had the highest purity and concentration and the best extraction efficiency. The CTAB method had the poorest extraction efficiency. Conclusion The extraction method of genomic DNA from the root extraction kit is simple, stable, and efficient. The extracted DNA is better and the purity and concentration of the extracted DNA are good.
