目的研究稀土元素镨对蚕豆种子萌发和早期生长的影响。方法应用0、2.5、10、40、160和640μg/ml6个质量浓度的PrCl_3培养蚕豆种子,3 d后统计发芽势,10 d后计算发芽率、发芽指数,并测量株高、根长、苗干重和根干重及幼叶和根部组织吲哚乙酸氧化酶(IAAO)的活性变化。结果各处理组均能促进蚕豆种子的萌发;2.5~40μg/ml处理组蚕豆幼苗的株高、根长、苗干重和根干重随Pr~(3+)浓度的升高而增大,且差异有统计学意义(P<0.05或P<0.01),而IAAO酶活性随Pr~(3+)浓度的升高而降低,10和40μg/ml处理组酶活性低于阴性对照组(P<0.05或P<0.01);当浓度≥160μg/ml时,幼苗的根长、苗干重和根干重均低于对照组,且差异有统计学意义(P<0.05或P<0.01),IAAO酶活性升高,且高于对照组(P<0.01)。IAAO活性与根尖细胞有丝分裂指数(MI)和根长呈负相关(r_(IAAO-MI)=-0.979,r_(IAAO-根长)=-0.993)。结论稀土镨离子可能诱导了蚕豆幼苗早期生长的Hormesis效应和IAAO酶活性的Hormesis效应。
Aim To study the effect of rare earth element praseodymium on the germination and early growth of Vicia faba. Methods Vicia faba seeds were cultured with PrCl_3 of 0, 2.5, 10, 40, 160 and 640 μg / ml. After 3 days, the germination potential was calculated. After 10 days, the germination rate and germination index were calculated. The plant height, Dry weight and root dry weight as well as the changes of indole acetic acid oxidase (IAAO) activities in young leaves and roots. Results The germination of Vicia faba L. seeds could be promoted by all treatment groups. The plant height, root length, dry weight and root dry weight of Vicia faba seedlings increased with the increase of Pr 3 + concentration in 2.5-40 μg / (P <0.05 or P <0.01), while the activity of IAAO enzyme decreased with the increasing of Pr 3 (3+) concentration. The enzyme activity of 10 and 40 μg / ml group was lower than that of negative control group <0.05 or P <0.01). When the concentration was more than 160μg / ml, the root length, dry weight and root dry weight of seedlings were all lower than those of the control group (P <0.05 or P <0.01) IAAO enzyme activity increased, and higher than the control group (P <0.01). IAAO activity was negatively correlated with mitotic index (MI) and root length (r_ (IAAO-MI) = -0.979, r_ (IAAO-root length) = -0.993). Conclusion Rare earth Pr 3+ ions may induce Hormesis effect and Hormesis effect of IAAO enzyme activity in early stage of broad bean seedling growth.
