本研究以盲肠结扎穿孔(CLP)方法制造腹腔感染模型,在CLP前(n=8)、CLP后4、8、12、24、48和96h(n=6)分别处杀动物,并留取血和肝脏标本。用差别PCR方法,建立了半定量检测肝脏白蛋白mRNA方法,初步观察到腹腔感染后4h,肝脏白蛋白mRNA开始下降,感染后48h,白蛋白mRNA下降到正常对照的27.2%,血浆白蛋白浓度与白蛋白mRNA密切相关(r=0.92,P<0.01),提示腹腔感染早期白蛋白基因表达受到明显抑制,腹腔感染时低白蛋白血症与肝脏白蛋白mRNA表达下降有关。
In this study, the model of intraperitoneal infection was made by cecal ligation and puncture (CLP). Animals were killed before CLP (n = 8), 4, 8, 12, 24, 48 and 96 h after CLP (n = 6) Blood and liver specimens. Hypoproteinemia was associated with the decrease of hepatic albumin mRNA expression .
