探讨了银杏内酯 B对体外血小板活化因子激活的大鼠中性白细胞某些功能 ,包括溶酶体酶的释放 ,超氧阴离子的产生以及细胞内游离钙浓度改变的影响。结果显示 :银杏内酯 B在 0 .5~ 10 0 μmol/ L 终浓度下 ,可使 β-葡萄糖苷酸酶释放量降低 44 .2 %~ 73.2 % ;在 0 .1~ 40μmol/ L终浓度下 ,可使超氧阴离子生量降低 2 7.4%~5 0 .0 % ;在 0 .1~ 10 0 μmol/ L 终浓度下 ,使 PAF和 f ML P刺激引起的细胞内游离钙浓度分别降低 13.9%~ 5 1.4%和 2 .2 %~ 36 .6 %。结果提示 :抑制溶酶体酶的释放、超氧阴离子生成以及降低细胞内游离钙浓度的升高可能与其抗炎作用有关。
The effects of ginkgolide B on in vitro platelet activating factor-activated rat neutrophils, including the release of lysosomal enzymes, production of superoxide anion, and changes in intracellular free calcium concentration, were investigated. The results showed that ginkgolide B could reduce the release of β-glucuronidase from 44. 2% to 73.2% at a final concentration of 0.5 to 100 μmol / L, and a final concentration of 0.1 to 40 μmol / L. In addition, the production of superoxide anion can be reduced by 27.4% to 50%; at a final concentration of 0.1 to 100 μmol/L, the intracellular free calcium concentration induced by stimulation of PAF and f MLP can be reduced, respectively. 13.9% to 5 1.4% and 2.2% to 36.6%. The results suggest that inhibition of lysosomal enzyme release, generation of superoxide anion, and reduction of intracellular free calcium concentration may be related to its anti-inflammatory effects.
