为研究旋毛虫病的血清学诊断技术,我们应用免疫酶染色试验法(IEST)检测家兔实验感染旋毛虫后抗体出现和消长情况,现将结果报道如下。 一、材料和方法 (一)抗原片制备:旋毛虫(Trichinella spiralis)肌内蚴(MSL)6000条接种大白鼠,3个月后剖杀取隔肌,咬肌置正已烷于乙醚加Co_2干冰速冻后贮于-20℃中待切片。冰冻切片组织厚度为4μm,每张玻片上贴8块组织片,贴片干燥后丙酮固定10分钟,pH7.4PBS洗涤2次,干燥后4℃贮存备用。
To study the serological diagnosis of trichinosis, we used the enzyme immunoassay method (IEST) to detect the appearance and growth of antibodies in rabbits infected with Trichinella. The results are reported as follows. Materials and methods (I) Preparation of antigens: 6000 rats were inoculated with Trichinella spiralis intramuscular larvae (MSL), and the septum was dissected 3 months later. Frozen dry ice stored at -20 ℃ to be sliced. The thickness of frozen sections was 4μm. Eight pieces of tissue pieces were attached to each slide. The dried pieces were fixed with acetone for 10 minutes, washed twice with pH 7.4 PBS, dried and stored at 4 ℃ for later use.
