目的探讨细胞内氯离子通道蛋白1(CLIC1)蛋白在结直肠正常黏膜、腺瘤及癌组织中的差异性表达及其与结直肠癌发生、发展和预后的关系。方法采用免疫组化组织芯片方法检测150例结直肠正常黏膜组织、62例结直肠腺瘤组织以及187例结直肠癌组织中的CLIC1蛋白表达情况,并分析CLIC1蛋白表达与结直肠癌临床病理因素及预后的关系。结果 CLIC1蛋白在结直肠正常黏膜组织(26.00%,39/150)、腺瘤组织(66.13%,41/62)和癌组织(82.89%,155/187)中的表达阳性率依次升高,之间的差异均有统计学意义(P<0.001)。CLIC1蛋白表达与结直肠癌的TNM分期有关(P=0.007),而与患者性别(P=0.553)、年龄(P=0.206)、肿瘤直径(P=0.185)、肿瘤分化程度(P=0.062)及肿瘤部位(P=0.598)均无关。CLIC1蛋白阳性表达者术后中位生存时间为80个月,CLIC1阴性表达者术后中位生存时间为111个月。log-rank检验表明,CLIC1蛋白阳性表达患者生存率(66.40%)明显低于阴性表达者(80.00%),P=0.031。结论 CLIC1蛋白表达与结直肠癌的发生、发展及预后有关,CLIC1可能是一个潜在的肿瘤标志物。
Objective To investigate the differential expression of intracellular chloride ion channel 1 (CLIC1) protein in normal colorectal mucosa, adenoma and carcinoma and its relationship with the occurrence, development and prognosis of colorectal cancer. Methods The expression of CLIC1 protein in 150 cases of colorectal normal mucosa, 62 cases of colorectal adenoma and 187 cases of colorectal cancer was detected by immunohistochemistry.
