目的应用乳酸-羟基乙酸共聚物(PLGA)制备载紫杉醇(PTX)PLGA微球,评价PTX-PLGA微球对荷瘤小鼠治疗效果。方法采用4周龄雌性BALB/c-nu裸小鼠建模,将卵巢癌Skov3细胞株注射于裸鼠背部一侧,制备裸小鼠皮下移植瘤。设空白对照组、生理盐水组、PLGA微球悬液组、PTX注射液组和PTX-PLGA微球悬液组。分别对成瘤后荷瘤鼠进行治疗、观察记录、绘制肿瘤生长曲线。对Skov3移植瘤裸小鼠瘤内直接注射药物并应用免疫组化法检测移植瘤微血管密度(MVD)、相关死亡促进因子(Bad)。结果给药6周后,PTX-PLGA微球组瘤内注射抑瘤效果明显大于其他各组(P<0.05),肿瘤体积明显减小,癌细胞增殖受到抑制。结论 PTX-PLGA微球瘤内直接注射对卵巢癌的治疗有明显体内抑瘤效果。
OBJECTIVE: To prepare paclitaxel (PTX) loaded PLGA microspheres by using lactic acid-glycolic acid copolymer (PLGA) to evaluate the therapeutic effect of PTX-PLGA microspheres on tumor-bearing mice. Methods 4-week-old female BALB / c-nu nude mice were used to model the ovarian cancer Skov3 cells on the back side of nude mice. The blank control group, saline group, PLGA microsphere suspension group, PTX injection group and PTX-PLGA microsphere suspension group. After the tumor-bearing mice were treated, observe the record, draw the tumor growth curve. The tumor was injected intratumorally into the tumor of Skov3 xenograft in nude mice, and the microvessel density (MVD) and the related death promoting factor (Bad) of the tumor were detected by immunohistochemistry. Results After 6 weeks of administration, the tumor inhibition effect of PTX-PLGA microspheres group was significantly greater than that of other groups (P <0.05). The tumor volume was significantly reduced and the proliferation of cancer cells was inhibited. Conclusion PTX-PLGA micro-intratumoral injection of ovarian cancer in vivo treatment has obvious anti-tumor effect.
