为了提高夏大豆导入外源DAN的效果,于1987~1988年在山东省农业科学院作物研究所大豆试验田设置了夏大豆观察圃。通过白花授粉后大豆子房的切片观察、探明了济南地区夏大豆的受精过程,确定了在济南地区气候条件下外源DNA导入大豆的适宜时机。本试验采用本所育成的“鲁豆4号”、“7517”和“7229”三个品种(系)。在开花盛期6:30左右挂牌标记花冠与花萼平齐的小花、7:00授粉(北京夏令时)。试验设15个处理,即授粉后1~10小时的每1小时和第12、24、32、48、72小时分别取样,每个取样处理固定10个子房,重复2次。
In order to improve the effect of summer soybean introduction of foreign DNA, a summer soybean observation garden was set up in 1987 ~ 1988 in soybean experimental field of Crop Research Institute of Shandong Academy of Agricultural Sciences. Through the observation of slicing of the ovary in the white pollination of soybean, the fertilization process of summer soybean in Jinan area was ascertained, and the suitable timing of foreign DNA introduction into soybean in the climate of Jinan was determined. In this experiment, we cultivated three varieties (lines) of “Lu Dou 4”, “7517” and “7229”. In the flowering period of about 6:30 listed marked Corolla flush with the calyx, 7:00 pollination (Beijing DST). In the experiment, 15 treatments were taken, that is, every 1 hour and 12, 24, 32, 48 and 72 hours after pollination were sampled from 1 to 10 hours, and 10 ovary was fixed for each sampling and repeated twice.
