目的 验证光感受器的细胞凋亡是否为实验性大鼠眼铁质沉着症视网膜损伤的机制之一。 方法 5 mg的消毒纯铁 (99.98% )颗粒经显微手术植入 32只实验组大鼠眼的玻璃体腔 ,体积相近的普通光学玻璃颗粒植入 10只对照组大鼠眼的玻璃体腔作对照。术后 1到 15 d的不同时间摘除眼球。用凋亡原位染色法 (Td T- mediated,d UTP- biotin nick- end labeling,TUNEL)观察受损视网膜神经元细胞核的改变 ;电泳观察视网膜细胞核 DNA核小体间降解所产生的阶梯状条带。 结果 实验组大鼠术后 2 d起在视网膜外颗粒层观察到 TUNEL 阳性细胞核 ,其数目随观察时间延长而增加 ,术后 15 d,外颗粒层仅有散在的正常细胞核残留 ;视网膜 DNA琼脂凝胶电泳观察到典型的阶梯条带。对照组大鼠则未见上述改变。 结论 光感受器细胞凋亡是实验性大鼠眼铁质沉着症早期视网膜损伤的机制之一
Objective To verify whether photoreceptor apoptosis is one of the mechanisms of retinal damage in experimental rat eye syndromes. Methods 5 mg of sterilized pure iron (99.98%) particles were implanted into the vitreous cavity of 32 experimental rats by microsurgery. Normal optical glass particles of similar volume were implanted into the vitreous cavity of 10 control rats. . Eyeballs were removed at different times from 1 to 15 days after operation. The changes of nucleus in injured retinal neurons were observed by TdT-dTPA (TdT-mediated dUTP-biotin nick-end labeling). The results of electrophoresis showed that the ladder- band. Results The number of TUNEL-positive nuclei was observed on the extra-granular layer in the experimental group 2 days after operation. The number of TUNEL-positive nuclei increased with the prolonged observation time. Only 15 days after operation, there were only scattered normal nuclei in the extragranular layer. Retinal DNA agar gel A typical ladder banding was observed by gel electrophoresis. The control group rats did not see the above changes.
