染色体的非整倍性和不稳定性是癌细胞中最普遍的两种异常情况。有丝分裂表达的控制失调、多极纺锤体的形成和胞质分裂缺陷都可能导致以上异常情况的出现。许多控制有丝分裂过程和染色体分离的细胞周期依赖蛋白激酶已经被鉴定出来,Nek2是其中的一种代表蛋白,它是细胞周期调控蛋白相关的丝/苏氨酸激酶家族中的一员。基于最新的研究阐述Nek2表达异常与癌症发生之间的关系,为更好地认识癌症发生机制提供新的思路。这不仅仅有利于肿瘤的早期诊断和预防,同时为抗癌新药,特别是为以Nek2为靶点的药物的研究提供新的理论指导。
Chromosomal aneuploidy and instability are the two most common abnormalities in cancer cells. Miscontrol of mitotic expression, formation of multipolar spindles, and defects in cytokinesis may lead to the above abnormalities. Many cyclin dependent protein kinases that control mitotic processes and chromosomal segregation have been identified, one of which is representative of Nek2, a member of the cyclin / threonine kinase family of cell cycle regulatory proteins. Based on the latest research, we elucidate the relationship between the abnormal expression of Nek2 and the occurrence of cancer and provide new ideas for better understanding of the mechanism of cancer. This is not only conducive to the early diagnosis and prevention of tumors, but also provides new theoretical guidance for anticancer drugs, especially for the research on Nek2-targeted drugs.
