【目的】研究人重组γ-干扰素(hrIFN-γ)和白细胞介素4(hrIL-4)对人早孕滋养层细胞PCNA和bcl-2表达的影响。【方法】采用人工流产方法提取绒毛膜组织,在体外培养条件下加入hrIFN-γ和hrIL-4,继续培养24 h,用倒置光显微镜和透射电镜观察绒毛膜组织形态学变化,应用免疫组织化学法检测绒毛膜组织滋养层细胞PCNA和bcl-2蛋白表达。【结果】hrIFN-γ处理的滋养层细胞PCNA和bcl-2表达均较对照组弱;hrIL-4处理的滋养层细胞PCNA和bcl-2表达较对照组增强。【结论】hrIFN-γ抑制滋养层细胞的增殖,hrIL-4促进滋养层细胞的增殖。
【Objective】 To investigate the effects of recombinant human IFN-γ and hrIL-4 on the expression of PCNA and bcl-2 in human trophoblast cells. 【Methods】 Human chorionic tissue was extracted by artificial abortion, hrIFN-γ and hrIL-4 were cultured in vitro and cultured for 24 h. Morphological changes of chorionic tissue were observed with inverted light microscope and transmission electron microscope. Immunohistochemistry Method was used to detect PCNA and bcl-2 protein expression in chorionic trophoblastic cells. 【Results】 The expressions of PCNA and bcl-2 in trophoblast cells treated with hrIFN-γ were weaker than those in control cells. The expressions of PCNA and bcl-2 in trophoblast cells treated with hrIL-4 were significantly higher than those in control cells. 【Conclusion】 hrIFN-γ inhibits the proliferation of trophoblast cells and hrIL-4 promotes the proliferation of trophoblast cells.
