目的 :采用改进的离子对高效液相色谱方法测定人血浆中二甲双胍的浓度。方法 :乙腈作为蛋白沉淀剂 ,除去酸化血浆样品中的蛋白 ,二氯甲烷进一步去杂质后 ,取上清液进样。流动相为甲醇 (5 6 % ) ,0 35mmol·L-1的十二烷基硫酸钠和 5mmol·L-1的磷酸盐缓冲液 (4 4 % ,pH 6 6 ) ,流速为 1 2mL·min-1,紫外检测波长 2 34nm。结果 :二甲双胍和内标 (哌仑西平 )的保留时间分别为 7 1和 4 8min ,标准曲线在 2 0~ 4 0 0 0mg·L-1浓度范围内线性关系良好 ,绝对回收率 >95 % ,检测限和定量限分别为 4和 10mg·L-1。天内和天间RSD <6 4 %。结论 :该方法适用于盐酸二甲双胍的治疗药物监测及临床药学研究
OBJECTIVE: To determine the concentration of metformin in human plasma by improved ion-pair high performance liquid chromatography. Methods: Acetonitrile was used as a protein precipitator to remove proteins from the acidified plasma sample. After dichloromethane was further decontaminated, the supernatant was injected. The mobile phase consisted of methanol (56%), sodium dodecyl sulfate at 0 35 mmol·L-1 and phosphate buffer (4 4%, pH 6 6) at 5 mmol·L-1 with a flow rate of 12 mL · min -1, UV detection wavelength of 2 34nm. RESULTS: The retention times of metformin and internal standard (pirenzepine) were 71 and 48 min, respectively. The calibration curve was linear over the concentration range of 20 ~ 40 mg / L and the absolute recovery was> 95% The limits of detection and quantification were 4 and 10 mg · L -1, respectively. RSD <6 4% in days and days. Conclusion: This method is suitable for the treatment of metformin hydrochloride drug monitoring and clinical pharmacy research
