目的:探讨补肾活血方对免疫性卵巢早衰(POF)小鼠的治疗作用。方法:以重组猪卵透明带(pZP4)蛋白两后脚掌和皮下多点注射免疫雌性BALB/c小鼠建立POF动物模型。小鼠随机分为正常组、模型组、雌二醇组和补肾活血方高、中、低剂量组(按生药量计分别为15.0,7.5,3.75 g.kg-1),分析ig 20 d后阴道脱落细胞涂片、外周血雌二醇(E2)浓度、血清抗透明带(pZP4)抗体变化及卵巢组织炎形态学观察。结果:补肾活血方高剂量组小鼠阴道涂片显示动情周期基本规则为6~7 d;高剂量组可明显升高小鼠血清E2浓度(P<0.01);高剂量组小鼠的血清抗pZP4抗体滴度显著下降(P<0.05);高剂量组小鼠初级卵泡和次级卵泡的数量显著增加(P<0.05)。结论:补肾活血方能改善卵巢的生殖内分泌功能和卵巢内各期卵泡和黄体的发育状况,并可在一定程度上抑制自身免疫早衰作用。
Objective: To investigate the therapeutic effect of Bushen Huoxue Recipe on immune premature ovarian failure (POF) mice. Methods: The animal model of POF was established by immunizing female BALB / c mice with the recombinant porcine zona pellucida (pZP4) protein two hind paws and subcutaneous injection. The mice were randomly divided into normal, model group, estradiol group and Bushen Huoxue Fang high, medium and low dose groups (according to the raw drug amount were 15.0,7.5,3.75 g.kg-1) Vaginal smear, estradiol (E2) concentration in the peripheral blood, serum anti-zona pellucida (pZP4) antibody and morphological observation of ovarian tissue. Results: The vaginal smear of high-dose Bushenhuoxue prescription showed that the basic rules of estrous cycle were 6-7 days. The high-dose high-dose group could significantly increase the serum E2 level (P <0.01) The titer of pZP4 antibody was significantly decreased (P <0.05). The number of primary follicles and secondary follicles in high dose group was significantly increased (P <0.05). Conclusion: Bushen Huoxue Decoction can improve ovarian reproductive and endocrine function and ovarian follicles and corpus luteum development status, and to a certain extent, inhibit the role of autoimmune premature aging.
