目的通过对国内临床分离5株阿萨希毛孢子菌(Trichosporon asahii,T.asahii)rRNA基因3个不同区域的RFLP分析,明确T.asahii是否具有种内基因多态性以及D1/D2I、TSI、GS1区在T.asahii基因多态性研究中的作用。方法提取T.asahii DNA,用特异引物PCR扩增D1/D2、ITS及IGS1区,分别用限制性内切酶HapⅡ、MboⅠ、AluⅠ和HhaⅠ对3个区域的PCR产物进行酶切、分析。结果 D1/D2I、TS区酶切结果在菌株间无差异;IGS1区HapⅡ酶切菌株BZP07003结果及HhaⅠ酶切菌株BZP07004结果与其他菌株不同。结论国内临床分离T.asahii具有种内基因多态性,在rRNA基因3个不同的区域中,IGS1区可用于T.asahii基因多态性的研究分析。
OBJECTIVE: To determine whether T.asahii has intraspecific genetic polymorphism and D1 / D2I, TSI, or not by analyzing the RFLP analysis of three different rRNA genes of Trichosporon asahii (T.asahii) , GS1 region in T.asahii gene polymorphism in the role. Methods The T.asahii DNA was extracted and the PCR products of D1 / D2, ITS and IGS1 were amplified by specific primers. Restriction endonucleases HapⅡ, MboⅠ, AluⅠ and HhaⅠ were used to digest the three PCR products. Results There was no difference between the isolates in D1 / D2I and TS. The results of HapⅡ digestion in strain IGS1 and BZP07004 in HhaⅠ were different from other strains. Conclusion The clinical isolates of T.asahii from China have intraspecific genetic polymorphism. IGS1 can be used for the analysis of T.asahii gene polymorphism in three different regions of rRNA gene.
