目的研究亚低温对脑缺血后纹状体ATP和多巴胺含量的影响并探讨二者之间的关系。方法沙土鼠双侧颈总动脉阻断脑缺血再灌注模型,脑缺血时间10min。动物分为假手术组、常温组、亚低温组,应用高效液相测定纹状体的ATP和多巴胺的含量。结果再灌注60min时,常温组纹状体ATP和多巴胺的含量为缺血前的64%和52%,亚低温组分别为缺血前的89%和80%,其含量均高于常温组(P<0.01)。再灌注60min时,亚低温组腺苷酸池(ATP+ADP+AMP)的含量也高于常温组(P<0.05)。结论亚低温可减少脑缺血后纹状体多巴胺的释放,这可能为其减轻脑缺血再灌注损伤的机制之一
Objective To investigate the effects of mild hypothermia on ATP and dopamine levels in striatum after cerebral ischemia and to explore their relationship. Methods The bilateral common carotid arteries of gerbils were occluded to establish the model of cerebral ischemia and reperfusion. The cerebral ischemia time was 10 min. The animals were divided into sham operation group, normal temperature group and mild hypothermia group. ATP and dopamine contents of striatum were determined by HPLC. Results The level of ATP and dopamine in the striatum at normal temperature were 64% and 52% before ischemia and 89% and 80% respectively in the mild hypothermia group compared with those in the normal temperature group P <0.01). The content of ATP + ADP + AMP in the mild hypothermia group was also higher than that in the normal temperature group at 60min after reperfusion (P <0.05). Conclusion Mild hypothermia can reduce the release of striatum dopamine after cerebral ischemia, which may be one of the mechanisms of reducing cerebral ischemia-reperfusion injury
