目的:探讨脂质体/水(脂/水)相中的分配,预测10-羟基喜树碱(HCPT)在大鼠小肠中吸收的可行性。方法:采取在体小肠吸收模型,研究不同pH条件下HCPT在大鼠小肠中的吸收;采取平衡透析法检测不同pH条件下药物在脂质体/水相中分配系数;探讨HCPT在脂质体/水相中分配与药物的在体吸收的相关性。结果:HCPT在脂质体/水相中的分配及在体小肠吸收随外界环境pH的不同而改变,两者相关性良好。结论:用脂质体/水相中的分配可以预测HCPT在大鼠小肠中的吸收。
Objective: To investigate the distribution of liposome / water (lipid / water) phase and predict the feasibility of 10-hydroxycamptothecin (HCPT) absorption in rat small intestine. Methods: The intestinal absorption model was adopted to study the absorption of HCPT in rat intestine under different pH conditions. The partition coefficient of drug in liposomes / water phase under different pH conditions was determined by equilibrium dialysis method. / Water phase distribution and drug absorption in the body. Results: The distribution of HCPT in the liposome / aqueous phase and the intestinal absorption in the body changed with the pH of the external environment, and the correlation between the two was good. Conclusion: The distribution of HCPT in the rat small intestine can be predicted by the distribution in the liposome / aqueous phase.
