目的建立乌饭树叶中主要活性成分乌饭树苷的含量测定方法。方法利用染色活性跟踪法,分离鉴定有效成分;采用高效液相色谱法测定其含量,色谱柱为Cosmosil-5C18-PAQ(4.6 mm×250 mm,5μm),流动相为0.3%乙酸溶液-乙腈梯度洗脱,流速1.0 ml/min,检测波长320 nm。结果乌饭树苷为乌饭树叶主要有效成分,其色谱分析的分离度良好,在0.052 08~0.520 8 g/L范围内线性良好,r值为0.999 8,加样回收率为98.61%。结论该有效成分测定方法灵敏度高,结果准确、简便,可为乌饭树叶的质量控制方法提供参考。
OBJECTIVE To establish a method for the determination of the main active ingredient of mellow glycoside in the leaves of Ulva pertusa. Methods The active ingredient was separated and identified by the method of dyeing activity tracking. The content was determined by high performance liquid chromatography with a Cosmosil-5C18-PAQ column (4.6 mm × 250 mm, 5 μm) and a mobile phase of 0.3% acetic acid in acetonitrile Elution, flow rate 1.0 ml / min, detection wavelength 320 nm. Results The result showed that the glycoside was the main active ingredient in the leaves of Radix angustifolia, and its chromatographic resolution was good. Its linearity was good in the range of 0.052 08-0.520 8 g / L with the r value of 0.999 8 and the recovery rate of 98.61%. Conclusion The method for determination of active ingredients has high sensitivity and accurate results. It can provide reference for the quality control of Radix.
