目的探讨重组苦瓜子抗HIV植物蛋白30(MAP30)对人胃癌BGC-823细胞增殖与凋亡的影响。方法将BGC-823细胞分为对照组(A组)和MAP30 10、20、30、40、50、60μg/ml实验组(分别为B1、B2、B3、B4、B5、B6组)。MTT、集落形成和裸鼠致瘤实验观察细胞增殖的抑制作用,光镜、电镜、荧光染色和流式细胞术检测细胞凋亡。结果与A组相比,MAP30呈剂量依赖性地抑制BGC-823细胞增殖,IC50为51.67μg/ml。B3组光镜下细胞变圆缩小,细胞膜发泡,可见凋亡小体;电镜下细胞表面微绒毛消失,细胞核和细胞质浓缩,核膜褶皱,染色质凝集。荧光染色和流式细胞术检测显示,B3组BGC-823细胞凋亡率较A组增加(P<0.05)。结论 MAP30可抑制BGC-823细胞增殖,并可诱导其凋亡。
Objective To investigate the effect of recombinant bitter melon seeds on the proliferation and apoptosis of human gastric cancer cell line BGC-823. Methods BGC-823 cells were divided into control group (group A) and MAP30 10, 20, 30, 40, 50 and 60 μg / ml experimental groups (B1, B2, B3, B4, B5 and B6 respectively). The inhibition of cell proliferation was observed by MTT, colony formation and tumorigenesis in nude mice. The apoptosis was detected by light microscopy, electron microscopy, fluorescent staining and flow cytometry. Results Compared with group A, MAP30 inhibited the proliferation of BGC-823 cells in a dose-dependent manner with IC50 of 51.67μg / ml. Under the light microscope, the cells in round group B3 turned round, the cell membrane became vacuolized, apoptotic bodies were observed, the microvilli disappeared, the nucleus and cytoplasm were condensed, the nucleus membrane folds and chromatin agglutination were observed under electron microscope. Fluorescence staining and flow cytometry showed that the apoptosis rate of BGC-823 cells in B3 group was higher than that in A group (P <0.05). Conclusion MAP30 can inhibit the proliferation of BGC-823 cells and induce its apoptosis.
