目的探讨含中链甘油三酯(medium chain triglycerides,MCT)的高脂饮食对血清和脂肪组织中脂联素、过氧化物酶增殖体激活受体γ(PPARγ)水平的影响。方法雄性Wistar大鼠,分别饲喂含MCT和长链甘油三酯(LCT)的高脂饲料,8w后病理检测右侧肾周脂肪细胞的大小。ELISA法检测血清中脂联素、TNF-α、胰岛素的水平。逆转录PCR测定脂肪组织中脂联素、PPARγ mRNA表达水平。结果MCT组大鼠体脂(BFA)、外周脂肪组织的细胞直径小于LCT组。MCT组的血清脂联素含量高于LCT组,两组间血清TNF-α无显著差异。MCT组外周脂肪组织中脂联素、PPARγ mRNA表达水平显著高于LCT组。结论MCT饮食可通过促进PPARγ表达,上调脂联素基因的表达,改善大鼠胰岛素抵抗。
Objective To investigate the effects of high-fat diet containing medium chain triglycerides (MCT) on adiponectin and peroxisome proliferator activated receptor γ (PPARγ) in serum and adipose tissue. Methods Male Wistar rats were fed with high-fat diet containing MCT and long-chain triglyceride (LCT), respectively. After 8 weeks, the size of right kidney peritoneal fat cells was detected. Serum adiponectin, TNF-α, insulin levels were measured by ELISA. Reverse transcription PCR was used to detect adiponectin and PPARγ mRNA expression in adipose tissue. Results The diameter of body fat (BFA) and peripheral adipose tissue in MCT group was smaller than that in LCT group. The level of serum adiponectin in MCT group was higher than that in LCT group, and there was no significant difference in serum TNF-α between the two groups. Adiponectin and PPARγ mRNA expression in peripheral adipose tissue of MCT group was significantly higher than that of LCT group. Conclusion MCT diet can improve the insulin resistance of rats by up-regulating the expression of PPARγ and up-regulating the expression of adiponectin.
